It depends. Actual combat between parties may be nice (if you can get enough players), but one on one fights don't sound like a good idea.
Different parties working to sabotage each other rock, though.

I mean separate parties. If you have players within one party split up and fight over something, it's not going to be good for continuing the campaign.
Players fighting one-on-one run into issues such as the fighter being unable to touch the flying wizard.
How could you have two parties fighting though. Wouldn't those be different games if they're two different groups of players.

They would. The idea is basically to make their paths cross. That said, it's pretty large-scale and hard to pull off.
A game isn't some transcendent plan, it's just how player interaction with the GM's world turn out.

Oh and to clarify my last comment since it might come off that way, I wasn't arguing but saying the danger is that you think PvP will work out, but then once it doesn't work out it's too late. So then the question becomes whether it's worth doing it anyway or risking it.

I always had a good-natured group that could take PvP well. In a way, player killing was often the highlight of a very good campaign.
