The Alter, Cu, Medea, Medusa, Emiya, and Gilgamesh are all variations which is different from the og cast. Only Herk and Hassan showed up as characters as their old selves and retained roles in the story and they were the characters with the least amount of development from the last games too

IDK if you can really call Cu Caster a variation in terms of his actual character, I can give you Cu Alter as that's literally Medb's stupid imaginary friend.

The new Casters are different. In that overall their character is the same but they have new roles. Cu instead of being a warrior now serves as a guide now that he is a druid and that was how he interacted with you. Caster Gil is the same now that he is a wise king. Nasu even once stated that he is different in that when he was writing Babylon he mentioned he had trouble with adding Gilgamesh as an ally since we already seen that in CCC, it's only when that he thought of writing Gil after he did his quest for immorality and returned that he was able to write.
Saber Alter is in the same category as Hassan and Herk, my mistake. The thing is as a character we never interacted with her much outside of an antagonist. Shinjuku fixed that.
Salem Medea was Circe. Medea did show up but she had a minor role
