Sometimes T-M gets away with the person being from a parallel dimension like Musashi or they had the luxury of being frozen in time when it was at least plausible for them to be an effeminate teenage boy like Artoria or they aspired to feminine beauty like da Vinci. It was addressed in Meiji Ishin event, though not exactly explained. She was born a woman and the clan elders didn't want her to succeed because of that, so they tried to support her younger brother Nobuyuki. Nobuyuki was a total failure though and he knew it, plus he loved his sister, so he decided the best thing he could do for her was to take the plotting elders down with him by staging crappy coups.
Basically, she's just a girl and that's all there is to it, but it did affect her life and reign.
