hero!kotomine is actually apparently full servant tier above Ciel, on the grounds that literally any jack ass from the street would go into the servant/aoko/kouma/DAA tier with 10 command spells worth of fuck you in the bank, IIRC.

No they disappear upon use, the bullet making contact and the consumption of the Command Seal happened basically simultaneously.
Although the characteristics of the Command Seals meant they were disposables which would disappear upon use

Kotomine had 11 Command Spells in total, not 16. I think I did the math once.
As to "Hero Kotomine", it was from the CM2 interview section. Originally, Nasu intended Kotomine to lay the beat-down on True Assassin (I think in comparison to the "Keys nailing True Assassin to the tree trick"). The plan was to have Kotomine explain in detail to Zouken as he facemelts Zouken, "how even a mere human can wound a Servant with the power of about 10 Command Spells".
As to Kotomine being close to Ciel level, it was explained that the usual Kotomine is no match to Ciel, but Zero Kotomine with his near-fanatism and Command Spells will defeat even Ciel.

mellon and I argued for a bit on whether or not bazett was physically on par with Ciel, or close to it, as of hollow. he said since mastering fragerach brought them to near parity, then by pure combat ability they were probably roughy equal, I disagreed and mentioned she probably got stronger after HA in addition to mastering fragerach or fragerach mastery increased her basic skill level, we decided to call a gentleman's disagreement and leave it at that, and McJon later mentioned there may have been something in the interview about how bazett was level 15 in HA and 30 when she mastered fragerach, or something like that, but couldnt reliably be sure if that was true
