He indeed only uses the Original in the story, and Notes's glossary does not mention Barrel Replica. So I will guess that this was only considered afterwards.
I can never 100% keep this straight, but do Crest Worms kill men faster than women, and do women have a higher chance of surviving and integrating them because the worms go for the uterus and feed off sexual arousal and the uterine wall?
Was Kingu in Babylonia actually a servant, or were they counted as an avenger only because of gameplay?

IIRC Kingu is just Enkidu's physical corpse revived with a new software, and the Class change to Avenger was him using Enkidu's shapeshifting to make him self similar to Gorgon's Saint Graph on the inside (but not on the outside so the sprite men wouldn't need to draw a new snake Enkidu for this fight).
Those are lust worms. They seem to be a subspecies of Crest worm that Zouken created.
Normal Crest worms don't do anything but act as an ankle monitor. The acting as circuits etc etc are all things Zouken added on.The floor of the secret room is wriggling around.
The worms must be active right now, as leech-like insects are writhing all over the floor.
Matou Shinji knows what these worms are.
These are lust worms that his grandfather, Matou Zouken, raises that love human blood, semen, and marrow.
Once swarmed by these lust worms, men will have their spine crushed, brain sucked, and will be made into a cripple.

Should I read or watch anything else besides Fate/Zero before checking out The Case Files of Lord El-Melloi II? I see a few volumes are fan-translated.
There are guest characters from other works like Kara no Kyoukai and FGO, but I don't think knowing too much about them is necessary to understand their roles in Case Files. At most you'll miss a few references.
Was Medea's subterranean city-temple something they invented for DEEN anime specifically, or it was "always supposed to be there, but was never shown in the VN" kind of situation?

It was just the subterranean cavern that is already in Fuyuki, that she had turned into her Workshop. Nothing strange about that since the entrance is right under the temple. If anything it's weird that she bothered with the temple to begin with instead of camping in the cavern the entire time, I guess Kojiro wouldn't have bothered to guard the cavern.
I'm pretty sure it's like, her dad died when she was really young and that's when her mom went all in on the Arthur stuff?
Why is the Second Owner of a land called that? Is there any meaning to it besides Nasu thought stringing together English words sounded cool?
