Question about the places where the holy grail appears
On several occasions we are told that the three places where the holy grail can appear are the Ryuudou temple, the Fuyuki church and Tohsaka's house. Why, if the grail can only appear in those three places, in Fate Zero it appears in middle of town?
Because there are four places where the leylines are good enough, and the park was the lowest on that list.

Yes. The four locations where the Grail, specifically the Lesser Grail can manifest are spots with appropriate leylines to connect to the Greater Grail for the Vessel to lie in wait. Any of them is viable. Once on the right spot and filled with enough Magical Energy from defeated Servants, the Vessel will turn into proper Lesser Grail, to provide the necessary energy to the Greater Grail and activate its wish-granting powers. That is the system behind Holy Grail in Fuyuki.

The place where the Lesser Grail manifested during the Fourth War has been destroyed thanks to its destruction by outside force, making it no longer suitable for the Grail to appear, ritually and functionally.
It is cyclicle and non random. Ryuudou, Rins house, Church, park, repeat

another question about UBW dialogue in the anime...
According to Rin in episode 22 Illya's heart is the holy grail but to activate it needs a vessel...When the holy grail (Illya's heart) is activated, does the vessel become the holy grail?
I ask this because in the same scene he says that Illya would become the holy grail but Gilgamesh didn't want that...

Yes, anyone with Magic Circuits can be Vessel that hosts the Holy Grail. It might be more accurate to say that the Vessel becomes part of the Lesser Grail that makes up Fuyuki's Holy Grail system.

What do you think about the possibility of a Tsukihime remake anime after the release of the second part of the game? are there any good chances?
Most likely to happen. They even have Mahoyo animated after all. And with that, the "Tsukihime anime doesn't exist" meme could finally be put to rest.
Though I prefer FHA got animated first before TsukiRE.
If they could animate hollow ataraxia. They could have done that before mohoyo..

But if they animate Shuumatsu Rokuon then Nasu will write another KnK story that they'll have to animate. They've clearly seen through his eternal loop scheme.

I think the movies had a great ending and they no longer see the need to adapt the new novel as they are happy with the ending of the last movie...

They already animated Mirai Fukuin as a side story after Epilogue, so animating Shuumatsu, another side story, isn't any different.
