    Well... they're certainly not incompatible, but Ayako hasn't shown a lot of romantic interesting in Shirou. I think she likes him, but his selflessness doesn't really appeal to her so terribly much. She does have that one scene where she sort of berates him for not living slightly for himself (I think it's an UBW scene, I can look it up if anyone wants), and I think that demonstrates her feelings quite well. She just isn't terribly interested in him. Not to mention she probably wouldn't want to ruin Sakura's chances (since Ayako is fond of her kouhai). If Shirou went after her, I think it could work out pretty well though, possibly even with Ayako bringing out a little selfishness, deeply buried.

    Hard to say though, it depends a lot on the specific scenario you have in mind.

    True, but I just don't think she's really all that interested in him. She likes him to be sure, but romantically? I don't quite think so.

    She could come to love him like that, certainly. Fairly easily even, but as of what I can tell, she doesn't. For that to happen, you'd need a fairly specific scenario. It would probably be a cute sort of love story though.
