If you mean all the extra features, I think those are only available in the "Grand Quest chapter". Outside of that, your Grand Saber only has an extra 1000 to their ATK/HP.
So, I can confirm that the Def Up buff applies when 3 cards that do not form a chain is done. Chain Error is not required. It's a really flexible way of boosting your Grand Servant's protection when combined with the NP Damage Cut effect. Just don't expect it to be functional in solo battles is all.

The flat +1k is great for lower rarity/HP-focused Servants in general. God knows how many more flat Atk bonuses Summer Gareth needs to finally have relevant damage.
EDIT: I know that this is controversial but I wish they gave Grands even more flat Atk and HP. Or heck, added extra Class Score nodes that grant flat Atk and HP.

Though that makes me think, if we get 5 each duel campaign because they want us to try things out or whatever, wonder if the play for these fights isn't to just grand whoever is actually best for these and then change it to whoever you actually want when you're done farming

It's weird because if you do the higher level farming quests you'll finish the board before the missions. The second highest level quest is like 40 runs for 1600 mats, and the highest level one is like 32. But you still need 50 runs total for the missions.
So the only Grand effect that works outside of Grand Duels is 1k ATK/HP?
1k is still a pretty solid bonus effect since it means doing more damage. But yeah, atm only Grand Duels allow for Grand effects (triple CE + Grand Score).

Devs just telling people to farm the Duel because it's the best bang-for-buck and even give insane amounts of Bond, ig. Or they're tempering run counts based on the "weakest" Servant to be Grand-ed: The 1* Servants.

Idk if all these grand duels are supposed to be piss-easy or if I'm just fortunate to have a level 120 Okita for the Saber duel and that I'll face difficulties in future duels.
I'm far from good at getting much out of combat in this game, I just click most skills and hope I do enough damage, but because of the lack of actual support units I felt like the highest difficulty fight was too far out of reach to even remotely 1t, so I guess it depends, because my Bedivere (only 100 tho, not 120) doesn't hit for shit against Musashi.
I'd like to throw my two cents on the speculation for the next Grand Duel opponents.
The first obvious candidates would be the ones already estabilished as Grands/Grand Candidates, so Archer Orion, Lancer Romulus-Quirinus, Riders Taigong Wang and Nemo, Assassins King Hassan and Tezcatlipoca, Casters Merlin, Solomon and Castoria, Berserker Tezcatlipoca.
That said, I don't think they'll stick with these ones, I'm expecting it will follow the same pattern as Musashi: existing Servant from the class with a revamped NP. I'll confine my guesses to 5* PHH Servants (with one exception) and try to exclude pseudos and divine spirits. Being a Pokémon fan has taught me that patterns are rarely kept, but given the limited data, I'll go with the Musashi framework (she's not from PHH, but she's a fringe case compared to other Lostbelt Servants).
Without further ado, let's go:
Archer: Arjuna is the best candidate from the available roster. He's a regular archer, not a shooter or thrower, and a top servant. Has many astras in myth to inspire a new NP. Gilgamesh is the top of the class in strength, but it was made a point in Solomon, I think, that he's not a Grand. Of course the Grand Duels aren't lore-compliant, but Arjuna being an actual archer puts him a tier above. Another candidate I considered was Artemis, due to her connection to Orion and the Archer class card being a woman, but she's a divine spirit and Musashi also isn't a knight. Atalanta too, also due to the class card, but she lacks the power.
Lancer: Scathach. Fan-favorite and another top servant. Regular spear-user. Has a second NP not used in-game. Enkidu would be a second option, but Scathach feels more true to the spirit of the class. Also, she is connected to the franchise's first Lancer, being a cool callback. Her illustrator being the same as Musashi might be a point against her, though.
Rider: either Ozymandias or Achilles. Some of the strongest and most famous members of their class. I'm partial to Ozy because Ramesseum Tentyris is a prime candidate for a better animation. Also, I have a Greek candidate for another class, and I think it would be cool if each duel had a representative from a different culture.
Assassin: this one I think it has to be King Hassan, even if he was already a Grand. No other assassin fits better than him. Kinda obvious, as he is the only natural candidate even in-lore. If I had to choose another option, it would be Semiramis, strictly due to power and significance as first poisoner.
Caster: Very hard one. If Goetia-Solomon will indeed be our anniversary servant, that would be my guess. From our current roster, I'm going with Adult Da Vinci. Caster will be the last duel to be unlocked, and given that the Grand Graph system was developed by Da Vinci Lily and that she is about to die, I feel like her assuming an adult form and fighting us would be a way to go out with a bang. Or maybe she dies and bring the adult one back after the fight.
Berserker: I feel like no PHH 5* Berserker has the gravitas of a Grand. So, I'm going with a 4*, Heracles. Is it his best class? No it is not, but he is the Greek hero famous for his fatal flaw being wrath. Also, the OG bahsahkah. For the Grand Duel, though, he would use the Heracles Megalos model from Agartha and have a new NP animation. My best 5* option would be Cú Alter, but with a Ríastrad NP to set him apart from Medb's OC husbando. If class changes are allowed, Berserker Enkidu would be a good fit, but he's supposed to be in mud doll form as a Berserker, and I don't think they will make new models for the Grand Duels.
I've no idea of the specific mechanism of the final 4 break bar Grand Duel Musashi fight, but I've seen people casually breeze through it 4T while in my experience Musashi seemingly has unlimited Guts
